Title: Isolation of Bioactive Constituents and Pharmacological Activities of Berberis Baluchistanica (Ahrendt)

Abstract: The traditional uses of medicinal plants in various ailments are mostly based on the indigenous knowledge which is transferred from generation to generation without scientific evidence. Berberis baluchistanica (ahrendt) has been used for the cure of internal injuries and ophthalmic problems. It is also used against cough and internal injury of livestock and human beings. Beside this it is effective for the removal of kidney stones, for curing toothache, ulcers, jaundice, without scientific evidence(s). In the present research work, Berberis baluchistanica hydromethanolic extract (BBME), and its sub-fractions were screened for various biological activities, to provide a valid scientific rationale to its ethno-medicinal uses...................................................................................................................................(can not be copied)... In case of Insecticidal activity maximum mortality (30%) was shown by chloroform fractions against Rhyzopertha dominica. ,While Tribolium castaneum was resistant to all the samples. The BBME showed promising in-vivo anti-pyretic, analgesic, and anti-inflammatory activities at test dose of 150 mg/kg. BBME also exhibited Gastroprotective effect and nephroprotective effect in the aspirin induced ulcerogenesis and gentamicin induce nephrotoxicity respectively. Furthermore, BBME exhibited significant hepatoprotective activity against Paracetamol induced hepatotoxicity in rabbits. Structures of the isolated compounds were elucidated using advanced spectroscopic and spectrometric techniques i.e. 1H-NMR, 13C-NMR, COSY, HMBC, NOESY, HSQC, IR, UV, EI-MS, HR-EI-MS, FAB-MS, HR-FAB-MS and [α]26D. Among the isolated compounds, berberisinol (1), was new natural product and 6 were previously reported but first time isolated from this plant. The other six are (2), berberine (3), palmatine (4), 8-oxoberberine (5), gallic acid (6), oleanolic acid and (7) β-sitosterol.
